Semaglutide is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist analogue. Structurally modified with an α-aminoisobutyric acid substitution at position 2 and a C-18 fatty diacid moiety attached via a γGlu-2xOEG spacer at Lys26, conferring resistance to DPP-4 cleavage and high albumin binding for extended half-life in research models.
